If you or any family member have ever had medullary thyroid carcinoma (MTC), or if you have been diagnosed with multiple endocrine neoplasia syndrome type 2 (MEN2) — a genetic disorder affecting multiple endocrine organs — you must not take Orforglipron. Additionally, while taking the medication, you should be vigilant for the following symptoms that may indicate a thyroid tumor: a lump or swelling in the neck, persistent hoarseness, difficulty swallowing, or unexplained shortness of breath. If you experience any of these signs, you must inform your doctor immediately.
Unlike many other GLP-1 agents (which usually require injection and have strict timing requirements relative to food intake), Orforglipron is currently the only oral GLP-1 tablet that can be taken at any time of the day. This means you do not need to schedule your medication intake around eating or drinking — whether you take it on an empty stomach, with a meal, or between meals does not affect the basic way the medication is used.
