Precautions for Orforglipron Use

Orforglipron is not a health supplement, nor is it a weight loss medication that you can buy and try on your own. It is a weight management drug that requires physician evaluation and long-term follow-up monitoring.

Orforglipron Treatment Guidelines

1. Before undergoing planned surgery or procedures requiring anesthesia or deep sedation, proactively inform all relevant medical personnel that you are taking this medication. Current data are insufficient to provide specific recommendations on preoperative dose adjustments, including whether fasting requirements need to be modified or if treatment should be temporarily discontinued.

2. Provide a complete medical history prior to initiating therapy. Conditions to inform your doctor about include: a history of pancreatic or kidney disease, severe liver disease, severe gastrointestinal issues (such as gastroparesis or digestive dysfunction), a history of diabetic retinopathy, as well as whether you are pregnant, planning to become pregnant, or breastfeeding.

3. Patients using insulin or sulfonylureas may need a dose reduction of these medications upon initiating Orforglipron, as determined by a physician.

4. Individuals taking oral contraceptives must switch to a non-oral contraceptive method or add a barrier method during the first 30 days after starting treatment, as well as for 30 days following each dose escalation.

5. Promptly report persistent or worsening nausea, vomiting, or diarrhea to your doctor—continuous vomiting and diarrhea can lead to dehydration, which may subsequently cause renal impairment.

Precautions During Orforglipron Treatment

The primary efficacy endpoint for Orforglipron is the mean percentage change in body weight after 72 weeks of treatment.

1. Reaching the target dose level takes time: titrating from 0.8 mg to 5.5 mg takes at least 60 days; each higher dose step requires at least an additional 30 days. Within the first three months, you are likely still in the dose escalation phase, and weight changes during this period are not representative of long-term results.

2. Consider stability as a positive outcome. Preventing further weight gain, reductions in waist circumference, and improvements in blood pressure and triglycerides are all documented benefits in the prescribing information, even if they do not manifest as rapid numerical weight loss.

3. Do not increase the dose on your own simply because you feel it is not working. Dose adjustments must be made by a physician; skipping dose tiers significantly increases the severity of gastrointestinal side effects, as the escalation period is when adverse reactions are most concentrated.

4. Do not stop taking the medication on your own. Weight management is a long-term treatment, and weight regain after discontinuation is a common reality with this class of drugs. Whether to continue, reduce, or stop treatment should be decided by your physician based on your weight, waist circumference, metabolic parameters, and overall health status.

Friendly Reminder: Orforglipron should be combined with a reduced-calorie diet and increased physical activity. Throughout treatment, patients should undergo standard lifestyle interventions, including guidance on a reduced-calorie diet and at least 150 minutes of physical activity per week.

Orforglipron Storage Instructions

Store Orforglipron at room temperature between 20°C to 25°C (68°F to 77°F), with temporary excursions permitted between 15°C to 30°C (59°F to 86°F).

Orforglipron is light-sensitive and should be kept in its original bottle and outer carton. Tighten the bottle cap immediately after each use. If the outer carton has been discarded, place the bottle in a light-protected area, such as a drawer or cabinet.

Do not remove the desiccant inside the bottle. Do not use past the expiration date printed on the bottle. The original packaging consists of 30 tablets per bottle equipped with a child-resistant closure.
