LuciDarax(Daraxonrasib) was approved by the U.S. FDA on August 26, 2026, under the brand name RASONQUE. It is the first oral targeted drug approved in the United States that directly targets RAS.
LuciDarax(Daraxonrasib) is an oral RAS GTPase family inhibitor and belongs to the class of "pan-RAS(ON) multi-selective inhibitors." It is used to treat adult metastatic pancreatic adenocarcinoma in patients who have received at least one prior systemic therapy or who are not suitable for multidrug combination systemic therapy.

LuciDarax(Daraxonrasib) binds to cyclophilin A, first forming a binary complex; this complex then binds to active-state RAS to form a ternary complex, thereby blocking the interaction between RAS and downstream effector molecules and promoting the hydrolysis of GTP to the inactive GDP-bound state.
In vitro studies show that it inhibits wild-type and mutant KRAS, NRAS, and HRAS, causes tumor growth inhibition and regression in RAS-dependent pancreatic adenocarcinoma models, and is associated with antitumor immunity.
Class: Targeted therapy; more precisely, a small-molecule inhibitor of the RAS family that acts on the "active state" of RAS (GTP-bound state, commonly abbreviated in the field as the ON state).
Targets: Wild-type and multiple mutant forms of KRAS, NRAS, and HRAS. It is not a single-target drug that hits only one mutation site.
Indication: Adult metastatic pancreatic adenocarcinoma after failure of at least one prior line of systemic therapy, or in patients who are not suitable for multidrug combination systemic therapy.
Dosage Form: Film-coated tablets, two strengths. 100 mg, light blue oval film-coated tablets; 150 mg, orange oval film-coated tablets.
Packaging: 30 tablets per bottle. The bottle contains one desiccant packet, and the cap is child-resistant.
Administration: Oral, once daily, 300 mg per dose, until disease progression or unacceptable toxicity.
Missed Dose: If more than 4 hours have passed since the scheduled dose time, skip the dose; do not take a double dose at one time.
Status: The world's first approved oral pan-RAS inhibitor; one of the first RAS(ON) multi-selective inhibitors to enter clinical trials.
Storage: 20°C to 25°C; brief excursions between 15°C and 30°C are permitted. Do not refrigerate. Do not store in the bathroom.
