During treatment with pralsetinib, it is imperative that you inform your healthcare provider of any prescription drugs, over-the-counter medications, vitamins/minerals, herbal products, and other supplements you are taking.
Before starting pralsetinib therapy, be sure to discuss with your healthcare provider if you are using any of the following medications:
1. Itraconazole (brand names: Sporanox, Tolsura), fluconazole, or voriconazole (brand name: Vfend) — these are antifungal medications used to treat fungal or yeast infections.
2. Verapamil (brand names: Verelan, Verelan PM) — a medication used to treat hypertension (high blood pressure) or chest pain.
3. Contraceptives, including birth control pills, patches, vaginal rings, implants, injections, or intrauterine devices (IUDs).
4. Numerous other medications may alter the blood concentration of pralsetinib. Disclose to your healthcare provider all medications you are currently taking or have taken recently.

Certain medications should not be used during or with specific types of food, as interactions may occur. Concurrent use of alcohol or tobacco with some medications may also trigger interactions. The interactions listed below are selected based on their potential significance and are not exhaustive.
Concurrent use of pralsetinib with grapefruit juice is generally not recommended, but it may be unavoidable in some cases. If co-administration is necessary, your doctor may adjust the dosage or frequency of pralsetinib, or provide specific instructions regarding the consumption of food, alcohol, or tobacco.
The presence of other medical conditions may affect the use of this medication. Be sure to inform your doctor if you have any other health problems, especially the following conditions, as pralsetinib may exacerbate them and requires cautious use:
1. Bleeding disorders.
2. Renal (kidney) disease.
3. Hepatic (liver) disease.
4. Pulmonary (lung) conditions (e.g., interstitial lung disease, pneumonia).
5. Uncontrolled hypertension.
6. Recent surgery.
