While certain medications should not be used together at all, in other cases, it may be necessary to use two different drugs simultaneously even if an interaction might occur.

What are the drug interactions of Lorlatinib?

In such cases, your doctor may wish to adjust the dosage or implement other preventive measures. It is particularly important to inform your doctor if you are taking any of the medications listed below while on this treatment. The interactions selected and listed below are based on their potential significance and are not exhaustive.

Concurrent use of this medicine with any of the following drugs is not recommended. Your doctor may decide not to treat you with this medicine or to change some of your other medications. These drugs include:

Apalutamide, Bacillus Calmette-Guérin (BCG) live vaccine, Carbamazepine, Dengue tetravalent live vaccine, Enzalutamide, Fosphenytoin, Ivosidenib, Lonafarnib, Lumacaftor, Mavacamten, Measles virus live vaccine, Mitotane, Mumps virus live vaccine, Phenytoin, Ranolazine, Rifampicin, Rifapentine, Rubella virus live vaccine, Smallpox and monkeypox vaccine, St. John’s Wort, Typhoid vaccine, Varicella virus live vaccine, and Yellow fever vaccine.

How should I use Lorlatinib?

Take this medicine orally. Follow the directions on your prescription label and take it at the same time each day. Do not cut, crush, or chew the tablets; swallow them whole. It can be taken with or without food. If it causes stomach discomfort, take it with food. Continue taking it unless instructed to stop by your medical team.

Do not take this medicine with grapefruit juice.

If this medicine is to be used for a child, consult your medical team first. Special monitoring may be required.

Overdose: If you suspect an overdose, contact a poison control center or emergency room immediately.

Note: This medicine is for your personal use only. Do not share it with others.

What side effects may Lorlatinib treatment cause?

Report the following side effects to your medical team as soon as possible:

1. Allergic reaction—skin rash, itching, hives, swelling of the face, lips, tongue, or throat.

2. Dry cough, shortness of breath, or difficulty breathing.

3. Changes in heart rhythm—rapid or irregular heartbeat, dizziness, feeling weak or lightheaded, chest pain, difficulty breathing.

4. Hyperglycemia—increased thirst or urination, unusual weakness or fatigue, blurred vision.

5. Increased blood pressure.

6. Changes in mood and behavior—anxiety, nervousness, confusion, hallucinations, irritability, hostility, suicidal or self-harming thoughts, worsening mood, feelings of depression.

7. Pain, tingling, or numbness in the hands or feet, muscle weakness, changes in vision, confusion or difficulty speaking, loss of balance or coordination, difficulty walking, seizures.
